    The DRX analyzers combine liquid handling, chip management and data capture for switchSENSE® measurements. Bespoke electronics manage the electrical actuation of biomolecules on the biochip and measurement of molecular dynamics via time-correlated single photon counting. The instrument also offers the proprietary electrical desorption methodology to adjust capture molecule densities. Instruments are designed for walk-away operation with automated dilution series / liquid handling, temperature managed environments and stability for long overnight / multi-day studies.

    The double color DRX2 analyzer offers the simultaneous measurement of two different species of probes on the same biochip electrode. Each sensor spot carries two nanolever sequences, one with a red tag, and one with a green. The instrument tracks the movement and position of the different nanolevers separately and simultaneously. This technology offers novel methods of investigation including same-spot controls, dual binding modelling, ideal for bispecifics and Proof of Action experiments with two nanolevers to play with, using one nanolever for capture and the second for detection.

    The switchSENSE® principle

    The principal functional elements of switchSENSE® are DNA nanolevers,
    which can be electrically actuated to oscillate at high frequencies.

    Turnkey conformational change analysis

    heliX® measures conformational changes in real-time by directly comparing the hydrodynamic friction (switching speed) of two molecules on two adjacent sensor spots.

    Conformational changes are expressed as %-friction-change, which works for any protein or nucleic acid state in almost any environment. Hence, molecular conformations can be investigated early in the discovery process, without previous knowledge about a sample.

    Simply define your initial state and measure change. Anything may serve as a reference, for instance:

    • bare DNA nanolevers without attached proteins
    • the protein conformation before small molecule binding
    • a protein mutant that cannot undergo conformational changes
    • a surrogate protein
    • a particular DNA or RNA fold
